Extradition bill protesters occupied Harcourt Road in Admiralty on Sunday evening, while many others gathered outside the police's headquarters in Wan Chai.
Marchers had earlier ignored a police demand for them to end their demonstration by Southorn Playground in Wan Chai, continuing on to Admiralty instead.
Shortly after 5pm, hundreds of protesters dressed in black and wearing face masks ran onto both sides of the busy Harcourt Road.
Bus and car drivers struggled to make their way through but eventually all lanes in both directions were totally blocked off.
One of the protesters, who gave his name as Jason and said he was a fresh graduate, told RTHK that nobody was sure of the next move.
"We don't know where we are going. We just followed the crowd and the crowd came to Admiralty. Maybe it's the Legislative Council [next] or something nearby, we don't know," he said.
Anticipating trouble in the Admiralty area, the police had already erected huge water barriers around the government's headquarters.
Meanwhile, a large number of protesters were gathered outside the police's headquarters in Wan Chai, shouting that officers are "shameful".
Barriers had also been put up around the complex which was besieged twice by extradition bill protesters last month.
Police wearing riot gear were watching protesters from inside the complex grounds.
